Inoculating Bacteria into a new jar

In a new bottle of Luria Broth, pipette 1 ml of your chosen bacteria and drop into the jar of Luria Broth.

To inoculate, place the luria broth bottle, now inoculated with bacteria into the shaking incubator.

Turn on the incubator switch on the left hand side. After it has loaded completely, press the knob until you can move screens to the “Programs” module

In the programs module, choose program #2 and press the button and choose “Run”, you should see a small man running on the bottom of the screen

The following are the steps for this program:

Step 1: 18 hr @ 37 degrees Celsius at 275 rpm

For 18 hours, the shaker will shake the bottle at 37 degrees Celsius. After 18 hours, it will stop shaking.

Step 2: 72 @ 4 degrees Celsius (take out ASAP)

After 18 hours, the bottle will stop shaking. Once it stops, you have 72 hours to remove the bottle and place it into the 4 degrees incubator, but it is best to remove as soon as possible.
